Salubrinal Activates Integrated Stress Response to Protect Against Doxorubicin-Induced Cardiotoxicity
What happened
Researchers found that the pharmacological ISR activator salubrinal alleviates doxorubicin-induced cardiotoxicity by activating ATF4-dependent pathways. This enhances glutathione (GSH) homeostasis and antioxidant capacity, suppressing apoptosis and preserving mitochondrial metabolism in H9c2 cardiomyocytes, C57BL/6 mice, and HL-1 cell models.
Why it matters
Doxorubicin is an effective chemotherapeutic agent limited by cardiotoxicity. This study highlights the integrated stress response as a vital survival mechanism and suggests that regulating antioxidant defenses via ISR activation and GSH homeostasis offers a promising therapeutic strategy for mitigating doxorubicin-induced cardiotoxicity.
